    Hyasen has developed sample direct amplification lysis buffer specifically for molecular POCT diagnostic scenarios. For direct amplification LAMP and direct amplification PCR systems, there is no need for nucleic acid extraction. The sample crude lysis buffer can be used for direct amplification, accurately detecting target genes, further shortening the sample detection time, and perfectly matching the molecular POCT application requirements. It is suitable for sample types such as nasal swabs and throat swabs, and can achieve the same results as conventional extraction methods without complex extraction operations. The pH of the sample lysis buffer is acidic, and the detection system needs to have a certain buffer. It is suitable for most PCR, RT-PCR, and LAMP fluorescence detection with pH buffer, but not for LAMP colorimetric detection without buffer.

    This product should be transported and stored at room temperature.

    Instructions

    1. Take 800μl of Sample Release Reagent into a 1.5 mL sampling tube for later use;
    2. Use a swab to collect nasal or pharyngeal swabs;
    • Nasal swab: Take out a sterile swab and put it into the nostril, slowly push it to a depth of about 1.5 cm, gently rotate it 4 times along the nasal mucosa, and stay for more than 15 seconds, then use the same swab to repeat the same operation on the other nasal cavity.
    • Pharyngeal swab: Take out a sterile swab and gently and quickly wipe the bilateral pharyngeal tonsils and posterior pharyngeal wall 3 times.
    1. Place the swab immediately after collecting the sample in the sampling tube. The swab should be rotated and mixed in the preservation solution for at least 30 seconds to ensure that the sample is fully eluted in the sampling tube.
    2. Incubate at room temperature (20 ~ 25℃) for 1 min to complete the lysis preparation.
    3. Both 25μl system LAMP and RT-LAMP are compatible with 10μL template addition for detection experiments.

    Notes

    1. The minimum amount of Sample Release Reagent for a single swab can be adjusted to 400μl, which can be adjusted according to the test requirements.
    2. After the sample is processed with Sample Release Reagent, it is recommended to conduct the next test as soon as possible, and the interval time should not exceed 1h.
