Product Overview
Velocut SfiI is a recombinant restriction endonuclease designed for rapid DNA digestion in molecular biology workflows. It is suitable for plasmid DNA, PCR products, and genomic DNA containing the SfiI recognition site. Under the recommended conditions with 1× reaction buffer at 50°C, digestion can typically be completed in 5–15 minutes.
The supplied color reaction buffer contains red and yellow tracking dyes, allowing digested DNA samples to be loaded directly onto agarose gels without adding a separate loading dye. In a 1% agarose gel, the red dye migrates similarly to approximately 2,500 bp double-stranded DNA, while the yellow dye migrates similarly to approximately 10 bp double-stranded DNA.
Velocut SfiI cannot be heat-inactivated. When enzyme removal is required before downstream applications, phenol-chloroform extraction or column purification is recommended. Cleavage of CpG-methylated or Dcm-methylated DNA may be inhibited, so the methylation status of the DNA template should be considered when planning digestion. Store at −20°C.
Product Features
- Rapid Digestion at 50°C: Supports DNA digestion in 5–15 minutes under the recommended reaction conditions, helping streamline restriction workflows involving plasmid DNA, PCR products, and genomic DNA containing the target recognition site.
- Direct Gel Loading: The supplied color reaction buffer contains red and yellow tracking dyes, enabling direct agarose gel loading and convenient monitoring of electrophoresis progress.
- Defined Methylation Compatibility: Cleavage of CpG-methylated or Dcm-methylated DNA may be inhibited. The enzyme cannot be heat-inactivated, so template methylation and post-digestion purification should be considered during workflow planning.
FAQ
Q1: What is the recommended reaction temperature for Velocut SfiI?
Velocut SfiI is typically used at 50°C with the recommended 1× reaction buffer. Digestion can generally be completed in 5–15 minutes under the specified conditions.
Q2: Can Velocut SfiI be heat-inactivated after digestion?
No. Velocut SfiI cannot be heat-inactivated. If enzyme removal is required before downstream applications, phenol-chloroform extraction or column purification is recommended.
Q3: Does DNA methylation affect Velocut SfiI activity?
Yes. Cleavage of CpG-methylated or Dcm-methylated DNA may be inhibited. Consider the methylation status of the DNA template when planning restriction digestion.
