Product Overview
HYF236 Velocut BclI is a recombinant restriction endonuclease developed for rapid DNA digestion in molecular biology workflows. It can be used to digest DNA containing the appropriate BclI recognition site, including plasmid DNA, PCR products, and genomic DNA.
Under the recommended reaction conditions, digestion can be completed in 5–15 minutes at 37°C. The enzyme is compatible with standard reaction buffer and color reaction buffer. The color reaction buffer contains red and yellow tracking dyes, allowing digested samples to be loaded directly onto an agarose gel without adding a separate loading dye. In a 1% agarose gel, the red dye migrates similarly to approximately 2,500 bp double-stranded DNA, while the yellow dye migrates similarly to approximately 10 bp double-stranded DNA.
DNA cleavage by HYF236 Velocut BclI may be inhibited by Dam or EcoBI methylation. The enzyme can be heat-inactivated at 80°C for 20 minutes.

FAQ
Q1: How long does HYF236 Velocut BclI take to digest DNA?
HYF236 Velocut BclI can complete digestion in 5–15 minutes at 37°C under the recommended reaction conditions. Actual performance may depend on the DNA substrate and reaction setup.
Q2: Can DNA methylation affect HYF236 Velocut BclI activity?
Yes. Cleavage may be inhibited when DNA is methylated by Dam or EcoBI methyltransferases. Consider the methylation status of your DNA when planning a digestion reaction.
Q3: How can HYF236 Velocut BclI be inactivated after digestion?
HYF236 Velocut BclI can be heat-inactivated by incubation at 80°C for 20 minutes.
