Product Overview
HYF249 Velocut DpnII is a recombinant restriction endonuclease designed for efficient digestion of DNA containing the appropriate DpnII recognition site. It is suitable for restriction digestion workflows involving plasmid DNA, PCR products, and genomic DNA. Under the recommended reaction conditions, digestion can typically be completed in 5–15 minutes at 37°C.
For convenient downstream analysis, a color reaction buffer containing red and yellow tracking dyes allows digestion reactions to be loaded directly onto an agarose gel without adding a separate loading dye. In a 1% agarose gel, the red dye migrates similarly to approximately 2,500 bp double-stranded DNA, while the yellow dye migrates similarly to approximately 10 bp double-stranded DNA.
Cleavage of Dam- or EcoBI-methylated DNA may be inhibited, so the methylation status of the DNA substrate should be considered when planning digestion. HYF249 Velocut DpnII can be heat-inactivated at 80°C for 20 minutes and should be stored at −20°C, with a shelf life of two years.
