Product Overview
Velocut StuI (HYF300) is a recombinant restriction endonuclease designed for efficient DNA digestion in molecular biology workflows. It supports the digestion of plasmid DNA, PCR products, and genomic DNA containing the specific recognition site, with reactions typically completed in 5–15 minutes at 37°C using the recommended 1× reaction buffer.
The supplied color reaction buffer contains red and yellow tracking dyes, allowing digestion products to be loaded directly onto an agarose gel without adding a separate loading dye. In a 1% agarose gel, the red dye migrates similarly to a 2,500 bp double-stranded DNA fragment, while the yellow dye migrates similarly to a 10 bp fragment. Velocut StuI can be heat-inactivated at 80°C for 20 minutes. Cleavage of Dcm-methylated DNA may be inhibited, so DNA methylation status should be considered when planning restriction digestion.

FAQ
Q1: How long does Velocut StuI take to digest DNA?
Velocut StuI typically completes digestion in 5–15 minutes at 37°C using the recommended 1× reaction buffer. Actual performance may depend on the DNA substrate and reaction setup.
Q2: Is Velocut StuI affected by DNA methylation?
Cleavage of Dcm-methylated DNA may be inhibited. Consider the methylation status of the DNA substrate when designing the digestion workflow.
Q3: Can Velocut StuI be heat-inactivated after digestion?
Yes. Velocut StuI can be heat-inactivated by incubating the reaction at 80°C for 20 minutes.
