Product Overview
HYF260 Velocut HhaI is a recombinant restriction endonuclease developed for rapid and efficient DNA digestion in molecular biology workflows. It is suitable for plasmid DNA, PCR products, and genomic DNA containing the appropriate recognition site.
Under recommended reaction conditions, Velocut HhaI can complete DNA digestion in 5–15 minutes at 37°C. The supplied color reaction buffer contains red and yellow tracking dyes, allowing digestion products to be loaded directly onto agarose gels without adding a separate loading dye. In a 1% agarose gel, the red dye migrates similarly to approximately 2,500 bp double-stranded DNA, while the yellow dye migrates similarly to approximately 10 bp double-stranded DNA.
Velocut HhaI can be heat-inactivated at 80°C for 20 minutes. However, cleavage of CpG-methylated DNA may be inhibited, so DNA methylation status should be considered when designing restriction digestion experiments.
Product Features
- Rapid DNA Digestion: Enables restriction digestion in 5–15 minutes at 37°C under recommended reaction conditions, helping streamline routine DNA processing.
- Convenient Gel Electrophoresis: The supplied color reaction buffer contains red and yellow tracking dyes, supporting direct loading of digestion products onto agarose gels.
- CpG Methylation Sensitivity: Cleavage of CpG-methylated DNA may be inhibited, providing an important consideration for experimental planning and DNA substrate selection.
FAQ
Q1: How long does Velocut HhaI take to digest DNA?
Velocut HhaI typically completes digestion in 5–15 minutes at 37°C when used with the recommended reaction buffer and conditions.
Q2: Can Velocut HhaI digest CpG-methylated DNA?
Cleavage of CpG-methylated DNA may be inhibited. If the DNA substrate is CpG-methylated, consider its methylation status when planning the digestion workflow.
Q3: Can Velocut HhaI be heat-inactivated after digestion?
Yes. Velocut HhaI can be heat-inactivated by incubation at 80°C for 20 minutes.
