Velocut XbaI - HYF219
Velocut XbaI Cat. No. HYF219

Product Overview

HYF219 XbaI Restriction Enzyme is designed for rapid and efficient DNA digestion in molecular biology workflows. Under recommended reaction conditions, it can digest DNA in 5–15 minutes, with an optimal reaction temperature of 37°C, helping streamline routine restriction digestion procedures.

HYF219 XbaI Restriction Enzyme is suitable for digesting plasmid DNA, genomic DNA, viral DNA, and PCR products containing the appropriate XbaI recognition site. Its compatibility with Universal Buffer and Color Buffer supports double or multiple restriction digestion in a single reaction tube when the selected enzymes have compatible reaction requirements.

For added convenience, Color Buffer contains a density reagent and red and yellow tracking dyes, allowing digestion reactions to be loaded directly onto an agarose gel without adding a separate loading dye. In a 1% agarose gel, the red dye migrates similarly to approximately 2.5 kb double-stranded DNA fragments, while the yellow dye migrates similarly to approximately 10 bp double-stranded DNA fragments.

The enzyme can be heat-inactivated at 80°C for 20 minutes. Note that DNA methylation by Dam methylase may block or impair cleavage. Although incubation for 3 hours does not show star activity under the stated conditions, longer incubation may increase the risk of nonspecific cleavage.

FAQ

Q1: What is the optimal reaction temperature for HYF219 XbaI Restriction Enzyme?

The optimal reaction temperature is 37°C. Under recommended reaction conditions, DNA digestion can typically be completed in 5–15 minutes.

Q2: Can DNA methylation affect HYF219 XbaI Restriction Enzyme activity?

Yes. DNA methylation by Dam methylase may block or impair cleavage at affected recognition sites. If digestion is incomplete, consider the methylation status of the DNA substrate when troubleshooting.

Q3: How can HYF219 XbaI Restriction Enzyme be inactivated after digestion?

The enzyme can be heat-inactivated by incubation at 80°C for 20 minutes. Under the stated conditions, a 3-hour incubation does not show star activity, but longer incubation may increase the risk of nonspecific cleavage. Follow the product protocol for your specific application.
