Velocut XhoI - HYF220
Velocut XhoI Cat. No. HYF220

Product Overview

HYF220 XhoI Restriction Enzyme is designed for rapid and efficient DNA digestion in molecular biology workflows. It enables DNA digestion in 5–15 minutes under recommended reaction conditions, with an optimal reaction temperature of 37°C, helping streamline routine restriction digestion procedures.

HYF220 XhoI Restriction Enzyme is suitable for digesting plasmid DNA, genomic DNA, viral DNA, and PCR products containing the appropriate XhoI recognition site. Its compatibility with Universal Buffer and Color Buffer supports double or multiple restriction digestion in a single reaction tube when the selected enzymes have compatible reaction requirements.

For convenient downstream analysis, Color Buffer contains a density reagent and red and yellow tracking dyes, allowing digestion reactions to be loaded directly onto an agarose gel without adding a separate loading dye. In a 1% agarose gel, the red dye migrates similarly to approximately 2.5 kb double-stranded DNA fragments, while the yellow dye migrates similarly to approximately 10 bp double-stranded DNA fragments.

HYF220 XhoI Restriction Enzyme can be heat-inactivated at 80°C for 20 minutes. Cleavage may be blocked or impaired when the substrate DNA is methylated by CpG methylase. Although 3-hour incubation does not show star activity under the stated conditions, longer incubation may increase the risk of nonspecific cleavage.

FAQ

Q1: What are the optimal reaction conditions for HYF220 XhoI Restriction Enzyme?

The optimal reaction temperature is 37°C. Under recommended reaction conditions, DNA digestion can typically be completed in 5–15 minutes. Follow the product protocol for the appropriate reaction setup.

Q2: Can DNA methylation affect HYF220 XhoI Restriction Enzyme activity?

Yes. DNA methylation by CpG methylase may block or impair cleavage. If digestion is incomplete, consider the methylation status of the substrate DNA when troubleshooting.

Q3: How can HYF220 XhoI Restriction Enzyme be inactivated after digestion?

The enzyme can be heat-inactivated by incubation at 80°C for 20 minutes. Under the stated conditions, a 3-hour incubation does not show star activity, but longer incubation may increase the risk of nonspecific cleavage.
