Product Overview
BsmBI (HYF212) is a Type IIS restriction endonuclease designed for sequence-directed DNA cleavage in molecular biology workflows. Unlike enzymes that cut within their recognition sequences, BsmBI cleaves outside its recognition site, making it useful for Golden Gate Assembly and other DNA assembly applications that require defined DNA ends.
BsmBI is used with the recommended 1× reaction buffer at 55°C. The supplied reaction buffer contains recombinant albumin to help improve enzyme stability. The enzyme can be heat-inactivated at 80°C for 5 minutes.
Cleavage of CpG-methylated or EcoBI-methylated DNA may be inhibited. The methylation status of the DNA substrate should therefore be considered when designing digestion or DNA assembly workflows.
Product Features
- Type IIS Restriction Activity: Recognizes a non-palindromic DNA sequence and cleaves outside the recognition site.
- Suitable for Golden Gate Assembly: Supports DNA assembly workflows that use Type IIS restriction enzymes to generate defined DNA ends.
- Defined Reaction and Inactivation Conditions: Operates at 55°C in the recommended reaction buffer and can be heat-inactivated at 80°C for 5 minutes. Cleavage of CpG- or EcoBI-methylated DNA may be inhibited.
FAQ
Q1: Why is BsmBI used in Golden Gate Assembly?
BsmBI is a Type IIS restriction endonuclease that cuts outside its recognition site. This property enables the generation of designed DNA ends for assembling DNA fragments in Golden Gate Assembly workflows.
Q2: What are the recommended reaction conditions for BsmBI?
Use the recommended 1× reaction buffer and incubate at 55°C. BsmBI can be heat-inactivated at 80°C for 5 minutes.
Q3: Can DNA methylation affect BsmBI digestion?
Yes. Cleavage of CpG-methylated or EcoBI-methylated DNA may be inhibited. Consider the methylation status of the DNA substrate when planning digestion and assembly experiments.
